Abstract

This study investigated the possible beneficial role of the coenzyme10 (CQ10) against Di (2-ethylhexyl) phthalate (DEHP)-induced neurobehavioral and neurotoxic impacts in juvenile rats. To do this, 40 male Sprague Dawley rats (3 weeks old) were alienated into four groups, each comprising 10 rats. The 1st group were orally given 0.5 mL of corn oil, the 2nd group were orally given 10 mg CQ10/kg.b.wt, the 3rd group were orally given 500 mg DEHP /kg.wt, and the 4th group were orally given CQ10 (10 mg /kg.b.wt) + DEHP (500 mg/kg.b.wt) for 35 days until the end of puberty (56th day postnatal day).
The rats were weighed once a week, and their dosages were adjusted accordingly. At the end of the dosing, depression, anxiety, spatial learning, memory, repetitive/stereotypic like behavior, and cognition were evaluated using the forced swimming test, tail suspension test, open field test, Morris water maze test, self-grooming assessment, and novel object recognition test, respectively. The brain contents of dopamine, gamma amino butyric acid (GABA), enzymatic and non-enzymatic antioxidant, malondialdehyde (MDA), interleukin-6, caspase-3, and tumor necrosis factor-alpha (TNF-α) were estimated. The histopathological changes and immunoexpressions of glial fibrillary acidic protein (GFAP) and brain-derived neurotrophic factor (BDNF) protein were followed in the the cerebral, hippocampal, and cerebellar brain tissues.
