Abstract

Title: Molecular characterization and evaluation of immunomodulatory activity of selected probiotic strains and their bioactive components in mice

Lactobacillus casei NM512 and Enterococcus faecium NM113 were previously isolated from the feces of breast-fed infants according to its immune modulatory properties. This work aims to isolate, identify, and characterize the bioactive surfactant component from the probiotic bacteria Lactobacillus casei NM512 and E. faecium NM113. The surfactants were isolated from the both probiotic strains and the yield production was ranging from 20 to 100 mg L-1 by using shaking 150 – 300 rpm. The surfactant was traced by thin layer chromatography, Fourier-transform infrared spectroscopy and nuclear magnetic resonance. The results revealed that the isolated surfactant constitute of sugar moieties and lipopeptide residues. The structure features of the surfactant were identified by 1H and 13C NMR analysis.
The surfactants was also assessed for its antioxidant ability targeting the DPPH free radical, cytotoxicity ability using MTT assay, anti-inflammatory ability using RAW264.7 macrophages and antimicrobial ability using the well agar diffusion method against five pathogens (C. albicans, St. aureus, P. aeruginosa, E. coli, H. pylori) beside that it tested as anti-inflammatory agent inside mices. The results conclude that the isolated surfactants have a great opportunity to be promising applications in probiotic and pharmaceutical preparations.
