Abstract

The present thesis aimed to analyse some drugs and their related compounds either in pure form, pharmaceutical preparations or biological aliquots. Such drugs are bromhexine, ambroxol, indomethacin, acemetacin, hydroxychloroquine, moxifloxacin, ofloxacin, terbutaline and bambuterol.Different analytical techniques were applied for determination of the studied compounds.A facile spectrophotometric and spectrofluorimetric technique were established for the assay of bromhexine and its active metabolite ambroxol using eosin Y. The studied drugs formed ion-pair complexes with eosin Y which improved their UV absorbance and enabled their spectrophotometric assay. While spectrofluorimetric assay depended on quenching the fluorescence of eosin Y upon reaction with the studied drugs.A green HPLC method for the assay of Acemetacin and its active metabolite indomethacin in dosage form and biological matrices were studied. The developed technique depended on using ethanol (green solvent) as an eluent to assay and separate the studied drugs in different matrices with UV detection. The method was applied in diverse dosage forms of the drugs and human urine.Sspectrofluorimetric analysis of two drug mixtures used in the treatment of hospital-acquired pneumonia in COVID-19 patients was carried out. Direct spectrofluorimetric assay of hydroxychloroquine and moxifloxacin was performed using 1%cetrimide and methanol as a solvent. The developed method could assay the two drugs in the presence of each other in synthetic mixtures and human plasma causing the sensitivity of the method to allow assay of the drugs down to their biological concentration.The assay of hydroxychloroquine in presence of ofloxacin was performed via the 1st derivative synchronous spectrofluorimetric method. The method assayed the two drugs in a laboratory-prepared synthetic mixture and in presence of each other in human plasma.Nuclear Magnetic Resonance Spectroscopy technique assayed terbutaline and its pro-drug bambuterol using deuterated water as a solvent and phloroglucinol internal standard. Optimization of instrumental conditions was established to allow sensitive and green analysis of the two drugs.The validation criteria of the developed methods were intensively studied. All the results were statistically analyzed and compared with those given by comparison methods. The suggested methods were validated following ICH guidelines. All the results were statistically analyzed and compared with those obtained by official or comparison methods. The results showed no significant difference assuring the accuracy of the methods.
