Abstract

This study aimed to evaluate the potential of SVF and PRP to regenerate spinal cord function after induction of multiple sclerosis in cats. Twenty-five adult Persian cats were allocated equally into five groups, group I (control negative), group II (control positive, induction demyelination by EB, then treated by PBS), group III received SVF, group IV received PRP, and group V received SVF + PRP. Cats were assessed for 28 days after receiving treatment, and outcome measures included functional, MRI, histological, immunohistochemistry, TEM, and gene expression assessments. Based on different evaluation analyses, The SVF and PRP group exceeded the other groups in terms of performance. According to our findings, the combination of SVF + PRP dramatically increased hindlimb motor activity and reduced lesion size in MRI imaging, Histopathological, and TEM; the SVF +PRP group exhibited apparently normal structure of gray and white matter and regular density and distribution of myelin around nerve fibers. Immunohistochemically, it was shown that myelin basic protein and Olig2 immunoreactivity had significantly increased, while Bax and glial fibrillary acidic protein immunoreactivity had significantly decreased. Additionally, the gene expression of neurotrophic factors was substantially highest in SVF + PRP.
