The present work aimed to investigate the prophylactic and the curative effects of Holothuria arenicola extract (HaE) in comparison with the standard chemical drug ranitidine (RAN), on indomethacin and cold stress -induced gastric ulceration (GU) in healthy rats. Sixty four rats were randomly divided into four main groups. Rats of the first (control) group administered distilled water orally, rats of the second group (ulcer group) administered single oral dose of indomethacin and exposed to cold stress, rats of the third (prophylactic) group were treated with HaE and /or RAN and then exposed to GU and rats of the fourth group (curative) were exposed firstly to GU then treated with HaE and/or RAN. The present work showed that administration of indomethacin with cold stress caused severe, hemorrhagic ulcers and histopathological alterations as well as lipid peroxidation, free radicals generation and DNA damage. The results demonstrated that pre or post-treatment with HaE and/or RAN exhibit significant ameliorative effect on nearly all studied parameters, significantly improved the histological architectures of rats stomach and decreased DNA degradation. Moreover, the study also demonstrated that the combination of HaE and ranitidine (especially in the pre-treated group) has more potent therapeutics effect on ulcer healing
