Abstract
The main objective of this study was isolation, identification and characterization of the most prevalent isolates of Riemerella anatipestifer and also preparation of Riemerella anatipestifer inactivated vaccine using two different adjuvants and studying the efficacy of this vaccine in ducks. Five isolates suspected to be Riemerella anatipestifer could be obtained through out morphological and biochemical diagnosis using API20E from 190 samples collected from ducklings with clinical symptoms of infectious serositis or carcasses of ducklings suspected to be died from infectious serositis. All five suspected Riemerella anatipestifer isolates from outbreaks, were morphologically and biochemically related to Riemerella anatipestifer. The results of isolation was confirmed using PCR and preparation of inactivated vaccine containing 4 local isolates was done using aluminium hydroxide gel adjuvant and montanide ISA206 adjuvant. The vaccine was injected into groups of ducks one week old s/c and control group left to control the infectious serositis disease in ducks and results of mortality and incidence of infection in all ducks after challenge test was recorded. Immune response was assessed using ELISA, PHA, IFAT and Challenge test and found that the inactivated vaccine had high immune efficiency and high protection against infection with contagious infectious serositis in vaccinated ducks
