Abstract

The present study was conducted to examine a probable renoprotection and cardio-protection of TLR4-IN-C34 against isoproterenol (ISO) induced acute kidney injury (AKI) and acute myocardial infarction (AMI) in rats. Four groups of rats were used: Control group; rats received normal saline 0.9% w/v, 1 ml/kg, SC, two consecutive doses for two days separated by 24 hours) ; ISO rats received ISO (100mg/Kg/day), 1 ml/kg of 10% w/v solution in normal saline ,S.C. for two consecutive doses 24 hours apart C341+ISO group ;rats received TLR4-IN-C34 in a dose of (1mg/Kg, 1 ml/kg of 0.1% w/v solution in water for injection, ip) for two days (each dose was injected intraperitoneally prior to each ISO injection by one hour ;C343+ISO group rats received TLR4-IN-C34 in a dose of (3mg/Kg, 1 ml/kg of 0.3% w/v solution in water for injection, ip) for two days (each dose was injected ntraperitoneally prior to each ISO injection by one hour). The development of AKI and AMI were assessed by employing renal and cardiac histopathological examination, assessment of kidney and cardiac function biomarkers including “serum creatinine, AST, LDH, CK-MB and CK “, inflammatory cytokines biomarkers” IL-8, IL-1β, IL-12 and troponin I, oxidative stress biomarkers “MAPK and MYD-88” and apoptotic markers “Bax, BCL2 and NF-κB”.TLR4-IN-C34 reserved normal levels serum creatinine and cardiac function biomarkers “AST, LDH, CK-MB and CK”, Also they exhibited anti-inflammatory effect, revealed by reduction in renal “IL-8, IL-1β, IL-12” and cardiac “IL-8, IL-1β, IL-12 and troponin I”, cardiac and renal oxidative stress in ISO injected rats was attenuated by TLR4-IN-C34, where they diminished level of “MAPK and MYD-88”, TLR4-IN-C34 has antiapoptotic effect as it decreases Bax and upregulates the anti-apoptotic BCL-2. And improved NF-κB, it also improved renal and cardiac section in histopathological study. from the conducted study, it could be concluded that TLR4-IN-C34 has protective effect against ISO induced-AMI and AKI possibly through antioxidant, anti-inflammatory and antiapoptotic mechanisms limiting renal and cardiac damage and dysfunction.
