Abstract

Aim: To investigate the influence of different wavelengths of LPT on the survival of hDPSCs submitted to substances leached from tooth bleaching gel. Methodology: hDPSCs;P6 were plated into sixteen plastic 96-well microtitration culture plates with a cell density of 1x103 cells/well. Then, they were exposed to a culture medium conditioned by a 35٪-hydrogen peroxide in-office tooth bleaching gel for 45 minutes except hDPSCs of positive control plate, after their seeding and incubating for 24 hours in a regular culture medium. All hDPSCswells exposed to the conditioned medium were irradiated once with laser therapy in contact mode using 450-nm, 650-nm or 976-nm. The cell viability of hDPSCs was assessed by the MTT reduction assay immediately and 24 hours after the
irradiation. Kruskal-Wallis statistics was carried out and followed by Mann-Whiteny test & unpaired samples t-test. Results: Each group’s statistical highest optical density value was recorded at 24-hours observation period. The positive control group showed the highest optical density values while the negative one presented the lowest values between all groups whether at 0 or 24-hour observation periods. Regarding the irradiated groups’ results, the phototherapy of infrared laser groups exhibited significantly higher optical density values than ired or bluelaser phototherapy in each assessment time. The optical density values of hDPSCs irradiated by red laser were significantly higher than those submitted to blue laser. Conclusions: The phototherapy of low-intensity red or infrared laser can enhance hDPSCs’ cell survival in response to substances leached from 35٪-hydrogen peroxide in-office tooth bleaching gel.
