Abstract

The present study was conducted to evaluate and compare between the effect of using parathyroid hormone 1-34 and zoledronic acid, separately or concomitantly on microarchitecture, alkaline phosphatase activity of alveolar process and bone mineral density of rats{u2019} mandible with glucocorticoid (GCs) induced osteoporosis via histology, histomorphometry, dual energy x-ray absorptiometry (DXA) scan, scanning electron microscope (SEM), bone morphomety, and alkaline phosphatase enzymatic assay analysis. A total of 146 adult female albino rats were used in the current study. All rats received a weekly dose 7 mg/kg body weight dexamethasone for 5 weeks. This was followed by scarification of 2 rats to assure osteoporosis induction. 144 rats were randomly distributed into four groups: Control group (36 animals): received daily S.C. injection of saline. Aclasta group (36 animals): received weekly S.C. injection of 7.5 {u1D707}g/kg body weight Aclasta. Forteo group (36 animals): received daily S.C. injection of 60 og/kg Forteo. Combination group (36 animals): received weekly S.C. injection of 7.5 {u1D707}g/kg body weight Aclasta plus daily S.C. injection of 60 og/kg Forteo. The animals were sacrificed at 2 and 4 weeks after initiation of drug administration. The mandible was retrieved, then divided into two halves. The hemi-mandibles were used as following: The right halves of mandibles of 96 rats (n = 96, 24 per group and 12 per time) were fixed in 10% calcium formol solution for 48 hours, before examining using DXA scanner for bone mineral density assessment. After that, hemi-mandibles were processed and the alveolar bone surrounding the roots of the first molar was examined through routine histological and histomorphometric evaluation
