Abstract

Moringa oleifera, a multi-purpose herbal plant used as an alternative for medicinal purposes worldwide, has demonstrated lipid-lowering and anti-obesity effects. Irisin/ fibronectin type III domain-containing 5 (FNDC5) protects against obesity by inducing browning in white adipose tissue. Therefor this study investigated the effects of Moringa oleifera aqueous extract (MAE) on irisin/FNDC5 expression with monitored some mechanisms of their action against HFD-induced obesity in rats. Obese-modeled rats were treated orally with either MAE (400 or 1000 mg/kg/day) or distilled water (vehicle) for 4 weeks.In Obese-modeled rats, MAE performed a dose-dependent marked anti-obesity, and anti-dyslipidemic activity as it succeeded significantly in up-regulation of FNDC-5 gene mRNA expression and serum irisin level coupled with reduction of the BMI and serum leptin level; it clearly increased serum PON1 activity and improved lipid profile values along with significant reduction in serum TNFα. Moreover, it showed anti-oxidant activity that was achieved from the marked improvement of the oxidative injury in cardio-hepatic tissues via significant decrease of MDA and NO values, associated with a significant elevation of CAT, SOD and GSH values. These findings were confirmed by the regeneration of the hepatic histopathological structure. In conclusion, MAE, as food supplement, could play a beneficial role in management of obesity and its complications; this could be exhibited through multiple pathways, mainly via up-regulation of FNDC5 gene expression and production of the soluble myokine ”irisin” that responsible for browning of white adipose tissue as well as increment of total body energy expenditure.
