Abstract

The work in this thesis is considered a further contribution to screening of natural lipase inhibitors from plants and natural products to serve as lead compounds for future drug discovery through in-vitroand in-silicostudies.
<.Introduction and review of literature: Pancreatic lipase is the principal lipolytic enzyme synthesized and secreted by the pancreas which is responsible for digestion of 50% -70% of of dietary fats.
< Therefore, reducing fat digestion through pancreatic lipase inhibition is an important approach for treatment of obesity.
< Orlistat is considered a potent inhibitor of lipase enzyme, as it inhibits the absorption of 30% of dietary fats and has been proved to be effective for the treatment of human obesity by the FDA.
< However, this powerful lipase inhibitor has some serious side effects like steatorrhea, fecal incontinence, flatulence, risks of vitamin deficiencies and liver diseases. Consequently, there is a need to explore safer alternative and complementary therapy to combat obesity.
<Plant based remedies generally have the advantage in being mild in their activity, in having a broad spectrum of possible multiple/synergetic effects and are considered to have less side effects than a totally synthetic based drug. Moreover, the use of plant-based resources as a potential platform for discovery and development of new drugs has become an important research field.
Also, lipase inhibitory activitiesof some plant extracts and isolated compounds from plantswere reported. .
