Abstract

Literature review revealed that many compounds containing thienopyrimidine nucleus exhibited potent activity against different types of cancer.
In addition, several thieno[3,2-d]pyrimidine derivatives fused or attached to other rings exhibited potent anti-breast cancer activity as targeted therapy with different mechanisms of action.
<These facts promoted the design and synthesis of new thieno[3,2-d]pyrimidine derivatives as potential cytotoxic agent against MCF-7 and MDA-MB-231 breast cancer cell lines.
<The designed compounds incorporated the core nucleus 7-phenylthieno[3,2-d]-pyrimidine moiety substituted at the 4-position with different substituents including flexible side chains, 5-membered heterocyclic or fused heterocyclic systems.
<Design and preparation of these targeted compounds, their spectral analysis as well as their evaluation for their cytotoxicity, is presented as the main text of the present thesis.
The present thesis comprises the following chapters:
Chapter 1:
Introduction It includes a literature survey about cancer especially breast cancer, its types and different chemotherapeutic strategies used for breast cancer treatment.
It also includes a review of thieno[3,2-d]pyrimidine derivatives possessing cytotoxic activity against breast cancer and different cancer cell lines.
Discussion of their structure activity relationship (SAR) studies wherever possible is not far of our attention.
<Chapter 2:
Aim of the work It discusses the aim of this study and the chemical and biological bases on which the target compounds have been designed.
It includes four schemes representing the synthetic routes for the preparation of the desired compounds.
Chapter 3:
Discussion It discusses in details the basic concepts of the experimental methods adopted for the synthesis of the starting, intermediates and final designed compounds referring to available literature.
It also investigates the structural elucidation of the synthesized compounds by different spectroscopic techniques (IR, 1H-NMR, 13C-NMR, in addition to MS).
