Abstract

T
he aim of the study was to evaluate the antibacterial effect of different irrigating protocols on the biofilm of enterococcus faecalis in root canal.
A total of 144 human single rooted teeth were mechanically prepared using protaper rotary nickel titanium system in the presence of 2.5% NoOCI until finishing file # (40/ 0.6). The tooth length will be standardized to 16 mm from the root apex to the coronal border.
Samples were divided into three groups according to final irrigation:
group I: Irrigation of teeth will be with biopure Mtad for 1min.
group II: Irrigation of teeth will be with 10ml Qmix 2in1 for 1min.
group III: Irrigation of teeth will be with 5ml of 2.25% NaOCL for 1 min followed by 5ml of 17%EDTA for 1 min then final flush with saline.
Each group will be further divided into Subgroup a: 16 teeth with passive ultrasonic activation (PUI). Subgroup b: 16 teeth without agitation. Subgroup c: 16 teeth with hydrodynamic agitation.

Bacterial Counting
By enumeration of colony-forming units (CFU) per 1 mL. All bacterial specimens will be vortexed for twenty seconds and 10-fold dilutions will be prepared in saline. Aliquots of 0.1 ml will be spread plated onto BHI agar plates, incubated at 37°C for 48 hours, and colony-forming units (CFU) per 1 mL to be enumerated.
Data were collected and statistically analysed.
All groups showed high antibacterial effect against the biofilm of enterococcus faecalis when used as a final irrigants, but Mtad gave the highest antibacterial effect .
The subgroup of passive ultrasonic irrigation with the three irigants showed higher antibacterial action than no agitation and hydrodynamic agitation.
CONCLUSION
1. Biopure Mtad has highest antibacterial action as a final flush irrigation against E.faecalis.
2. Passive ultrasonic activation increases the antibacterial effect on all used irrigation solutions.
3. Hydrodynamic agitation with gutta percha is an easy, cheap and simple but effective way to activate the irrigants in the root canal.
4. Complete eradication of the biofilm of E.faecalis from root canal still a challenge.
