Abstract

Regarding that AD is a complex multifactorial disease involving many pathological processes, thus, treatment with multifunctional intervention like VLP constitutes a viable therapeutic approach. VLP extract displayed several pharmacological effects including antioxidative, metal chelation, anti-amyloid and anti-inflammatory properties.
Biochemically, the AD-rats also showed a significant decline in ACh levels, as well as, an increase in AChE activity. VLP administration showed marked amelioration in neurocognitive abilities, including; downregulation of AChE and increased ACh and DA, decreased tHcy, and restored antioxidant capacities level more effectively rather than RIVA, as well as, it reduced oxidative stress, that were observed in all treated groups versus AD-rats. VLP showed therapeutic effects against neurodegenerative-mediated changes by inhibiting neuropathological processes by supporting regulation of neurovascular system; this involves modulation of oxidative imbalance in brain, inhibiting Aβ-plaque formation, ameliorating impaired cholinergic system, which finally results in enhancement of neurocognitive function. This potential impact was observed in both VLP negative control and AD-VLP rats. Herein, VLP attenuated the neurocognitive decline and decreased inflammatory-oxidative process markers more effectively rather than RIVA, which have been proved from the biochemical and behavioural assessment.
In the present study, the modulation occurred in the oxidant/inflammatory/lipid metabolism/cholinergic profile markers in AD-rats treated by VLP support the antioxidant properties of the Egyptian plant Vitis vinifera and hence its usefulness in the treatment and management of AD, and indicating that grape polyphenols have neuroprotective and anti-amnesic effects against AlCl3-induced cerebral damages and cognitive decline.
