Abstract

Being a major social and medical problem, sexual
dysfunction may occur in both sexes. Sildenafil citrate
Viagra is selective inhibitor of phosphodiestrase 5 which is
present in both vascular tissue and brain, mainly
cerebellum. Inhibition of phosphodiestrase 5, causes
increased blood flow in the ischemic areas. Cerebellum
contributes to coordination of movement and some
cognitive functions as attention and language. This work
was to clarify the histopathological effects of sildenafil
citrate on the cerebellar cortex of adult male albino rats.24
adult male albino rats weighing 200-250 gm were divided
into four groups (6 rats were included in each group). The
rats of treated groups (II, III and IV) received respectively,
0.25mg/kg, 0.70mg/kg and 1.43mg/kg body weight of
sildenafil citrate dissolved in distilled water daily for 30
days. And the first group was the control group received
equal volume of distilled water. At the end of experiment,
cerebellar cortices were processed for light microscopy,
GFAB immunostaining and transmission electron
microscopic examination. On examination of low dose
treated, cerebellar cortical architecture was preserved, but
partially congested and dilated vessels were noted in all layers of cerebellar cortex and also in between folia. In rats
treated with 0.7mg/kg, most of purkinge cells appeared
irregular, deeply stained with ill-defined nuclei, In high
dose treated group, diffuse vacuolations, shrunken deeply
stained purkinje cells with pyknotic nuclei were observed.
An increase in GFAP immunoreaction was noted in all
layers in both intermediate and high doses treated groups.
from the present study it was concluded that low dose of
sildenafil citrate improved the blood flow with resulting in
hyperemia of cerebellar cortex of adult male albino rats. On
the contrary high doses of sildenafil citrate resulted in
neurodegeneration and reactive astrocytosis mainly in the
purkinje cell layer of cerebellar cortex of adult male albino
rats .
