Abstract

The study presented in this thesis include development and the combination between some techniques and technologies such as Analytical chemistry, Biochemistry, Photochemistry and Nanotechnology for the determination of some active drugs components (group of Tetracyclines) and human enzymes such as alpha fucosidase.
The thesis is divided into three chapters:
Chapter one:-
Including a general introduction on lanthanides, optical sensors and other methods used for determination of alpha fucosidase Enzyme.
Chapter two:-
Studying the five optical sensor Eu3+-Tetracyclines Complexes for assessment of alpha –fucosidase enzyme activity in human liver and select the best one at optimum condition suitable for the determination of alpha fucosidase activity in some real serum samples. The optimal experimental conditions such as solvent, pH and molar ratio corresponding to maximum luminescence intensity were investigated on 5-Complexes of Europium -Tetracyclines.
Chapter three:-
Describes novel, simple, sensitive and selective spectrofluorimetric method for the determination of Tetracyclines in pharmaceutical tablets, serum samples using optical sensor Eu3+-Tetracyclines. The product of Reaction Alpha fucosidase with its substrate 2CNP can remarkably quench the luminescence intensity of Tetracyclines complex in ACN at pH 8.4 and λex= 410nm( energy transfer from Probe to enzyme)The quenching of luminescence intensity peak of Eu3+-Tetracyclines complex at λem =618 nm is proportional to the concentration of 2-CNP and Eu3+-Tetracyclines complex nano optical sensor as a result that suggested profitable application as a simple nano optical sensor for Tetracyclines assessment. The dynamic ranges found for the determination of Tetracyclines concentration are (1.25×10-9to 1.0×10-5) mol L-1.The limit of detection (LOD) and Quantitation limit of detection (LOQ) are (8.3x10-10 and 2.5x 10-9)mol L-1 respectively. Under optimal conditions, the luminescence intensity of nano optical sensor Eu3+-Tetracyclines is inversely proportional to the concentration of 2-CNP in range (5.5 x10-10 to 1.05 x 10-6) mol L−1, (r= 0.992).The detection limit (LOD) (7.6 x 10-11mol L-1) and Quantitation limit of detection (LOQ) are (2.3x10-10molL-1 respectively. This method is used for the determination of alpha fucosidase in human liver.
Chapter Four:
Gatifloxacin concentration was determined by the complexation with the Tb3+ ion doped in sol-gel matrix and the possibility of the enhancement of the Tb3+ luminescence sensitized by (GFX) was established and investigated.
