Abstract

The present study was carried out in Agriculture Research Center Giza, Egypt to investigate: the effect of diet containing different ratios of sweet and bitter lupine on serum glucose, lipid profile and liver and kidney functions in induced diabetic rats.
Seventy male albino rats of Sprague- Dawley strain weighting (200+ 10g) were divided into ten groups; each group included 7 rats caged individually. Food and water were provided adlibitum; all rats were individually weighted before experiment and housed individually in cylindrical wire cages. The rats were divided into two main groups as the following: The first main group Will be fed on standard diet ”as a control negative group, The second main group : (63 rats) were injection with streptozotocin [ STZ, 45 mg/kg ] for hyperglycemic rats were divided into 9 subgroups according to the following: (7 rats) for each subgroup:
The first subgroup: will be fed on standard diet (SD) only (control positive group).
The second subgroup: will be fed on (SD) + 5% sweet lupine
The third subgroup: will be fed on (SD) + 10% sweet lupine .
The fourth subgroup: will be fed on (SD) + 5% bitter lupine .
The fifth subgroup :will be fed on (SD)+ 10% bitter lupine .
The sixth subgroup: will be fed on (SD)+ 5% removed bitterness sweet lupine .
The seventh subgroup: will be fed on (SD) + 10% removed
bitterness sweet lupine .
The eights subgroup: will be fed on (SD) + 5% removed bitterness bitter lupine .
The nineth subgroup (7 rats): will be fed on (SD) + 10%
removed bitterness bitter lupine The experimental period (6 weeks), all rats were individually weighed at the beginning, twice weekly in the first 4 weeks, then once / week in second two weeks. Feed intake was recorded every day. At the end of the experimental period, rats were fasted over night then anaesthetized before sacrifice and blood samples were collected from each rat and received into heparinized clean tubes then centrifuged for 10 min. At 3000 R.P.M to separate plasma. It was carefully seaprated into dry clean Wasserman cuvets by using pasteyr pipette and kept frozen till analysis at – 20C.
After rats anaesthetized heart, liver, pancreas, and spleen of each rat were removed and washed in saline (0.9% Nacl) then weighted immediately. Then kept in 10% formalin for the purpose of histological study.
Chemical analyzed of (sweet and bitter lupine ) before and after remove of bitterness, and analysis of total phenolics, which was the highest rate in the bitter lupine (111mg/g) and antioxidants, which was the highest percentage in bitter lupine after remove of bitterness (20.7mg/g
) ,amino acid analysis of four samples Which showed that glutamine found the largest ratios, especially in bitter lupine and fatty acid analysis Of four samples Which showed that Oleic acid Is present higher than the rest of fatty acids especially in sweet lupine .
Phenolic compounds (HPLC) of (sweet and bitter lupine ) before and after remove of bitterness of four samples which showed that e-vanillic is the most important phenolic acids that found in bitter lupine by 285.274 mg/g.
