Abstract

The work presented in this thesis included a Phytochemical and biological investigation of some Egyptian Senecio species (Senecio vulgaris, Senecio desfontainei, Senecio cineraria and Senecio petasitis).
This thesis consists of four parts:
Part I
PHYTOCHEMICAL INVESTIGATION OF SENECIO VULGARIS L.
Chapter I:
Isolation of four compounds from the whole plant of S.vulgaris L.
Chapter II:
Characterization of four compounds isolated from S.vulgaris L
The investigation of ethyl acetate and chloroform fractions resulted in the isolation of four compounds; a flavonoid and a coumarin from the former and two alkaloids from the later. Structural elucidation of the isolated compounds was carried out using different spectroscopic techniques (UV, 1D-NMR and 2D-NMR).
Chapter III: Determination of the ratio between senecionine and seneciphylline in S.vulgaris L. using 1H-NMR data
This part includes determination of the ratio of two common alkaloids in genus Senecio; senecionine and seneciphylline, as they were isolated as a mixture from S.vulgaris. Determination of the ratio was by comparing the integration of the characteristic1H-NMR peaks of each alkaloid H-13’ of senecionine was observed at δH 0.90 (3H, d, J=5 Hz), integration 3.05. On the other hand, H-13’a of seneciphylline was observed at δH 5.34 (2H, br.s), integration 2.82 and H-13’b at δH 5.00 (2H, m), integration 2.39 which means that the ratio of senecionine to seneciphylline is 1:2
Part II
COMPARATIVE STUDY OF THE ALKALOIDAL CONTENTS OF FOUR EGYPTIAN SENECIO SPECIES
This part includes the comparison of the alkaloids bearing chloroform fractions of four Egyptian Senecio species (S.vulgaris, S.desfontainei, S.cineraria and S.petasitis). This was done by TLC technique followed by spraying of the TLC plate with Dragendorff’s reagent, capturing an image of the plate, and then image analysis using SorbfilTLC program.
The results of the study showed that the order of the alkaloidal content from the highest to the lowest was as following; S.cineraria, S.desfontainei, S.vulgaris and S.petasitis.
Part III
BIOLOGICAL INVESTIGATION OF SOME EGYPTIAN SENECIO SPECIES
CHAPTER I:
In -vitro cytotoxicity screening of some Egyptian Senecio species.
CHAPTER II:
In-vitro screening of the free radical scavenging activity of some Egyptian Senecio species.
CHAPTER III:
Antibacterial and antifungal activities screening of some Egyptian Senecio species.
This part evaluated and compared cytotoxic activity against colorectal carcinoma cell lines (HCT-116), free radical scavenging activity as well as antibacterial and antifungal activity of different fractions of the four Egyptian Senecio species.
