Abstract

Infectious bronchitis virus is highly contagious disease of chicken constitutes one of the most common and difficult diseases to control. 13 chicken flocks suspected to be infected with IBV. In Ismailia, Sharkia & Dumyat province were selected for the study during 2011 and 2012. Pooled samples were inoculated in 9 day SPF embroynated chicken eggs, three successive passage by allantoic sac route. Inoculated eggs are examined for early embryonic deaths, curling, dwarfing and stunting of embroys and urate deposition in kidneys and ureters . Results of HI indicated that, 13 isolates were positive to IBV and negative for AIV. Six isolates are positive to NDV. Six chicken flocks are infected with IBV and simultaneously infected with NDV (Mixed infection). However only seven flocks are infected with IBV (single infection). Gel electrophoretic pattern of ORF1a gene of IBV isolated from 13 isolates shows 143 bp product. However, F gene of NDV are amplified at 320 bp in 6 samples only.Unfortunately S1 gene of IBV, H5 and H9 avian influenza gene could not generate PCR products at 1230 bp , 808 bp and 456 bp respectively. Another three forward and three reverse primers with several combination between them ( 6 trials ) are used but the electrophoretic pattern didn’t show any PCR products.
