Abstract

This work aimed to investigate the effect of nanoparticle calcium phosphate as adjuvant compared to aluminium phosphate (alum) asadjuvant. Histopathological changes and physiological changes post immunization with both calcium phosphate nanoparticles and Alum were investigated. Antibody, IFN-y and IL¬10 levels were time dependent. Calcium phosphate with ~¬probiolactone (CAP-BPL)-Rabies vaccinated group showed the highest results compared to that elicited post 1M and SC immunization with Alum-BPL-Rabies adsorbed vaccine and plain vaccine when evaluated using ELISA. Subcutaneous route of administration was an appropriate delivery mode for adsorbed vaccine models. The effects of calcium phosphate and aluminium phosphate adjuvants were examined in mice for liver, kidney and spleen toxicity. Results recorded that detected tissue irritation due to calcium phosphate and aluminium phosphate was ended on the 4th and 8th week post vaccination, respectively.In concolusion, calcium phosphate nanopm1icles (CPN) is a promising immune stimulating agent with a long term release of antibodies and safety measures compared with the currently used alum. Accordingly, it is recommended that CPN as adjuvant be used subcutaneously.
