Abstract

Lipoic acid is a potent antioxidant, while tadalafil is one of PDE-5 inhibitors. This study was designed to investigate the effect of -lipoic acid and tadalafil on endothelial function in streptozotocin induced diabetic rats. Type II diabetes was induced by low dose STZ 35mg/kg i.v. The results of the present study revealed that lipoic acid (50mg/kg/day i.p.) for 4 weeks produced significant decrease in fasting plasma glucose level, significant increase in fasting plasma insulin level, and significant increase in fasting plasma nitric oxide level. On the other hand tadalafil (2mg/kg/day p.o.) for 4 weeks produced insignificant change in fasting plasma glucose and insulin levels. It produced significant increase in fasting plasma nitric oxide level. Both -lipoic acid and tadalafil produced significant decrease in the vascular contractility of the perfused aortic rings to phenylephrine and increased the relaxation response induced to acetylcholine.
