Abstract

The p r e s e n t s t u d y aimed a t e v a l u a t i o n 6 the schistosomicidal a c t i v i t y of f o u r newly s y n t h e s i z e d n o n - a n t i m o n i a l compounds o f t h e g e n e r i c name c h a l c o n e s ; n a m e l y : c h a l c o n e 1,2,3 and 4. The i n t e r a c t i o n o f t h e s e compounds as compared w i t h p r a z i q u a n t e l , t h e d r u g o f c h o i c e u s e d i n Egypt f o r t h e mass c h e m o t h e r a p e u t i c c o n t r o l of s c h i s t o s o m i a s i s , w a s i n v e s t i g a t e d a t b o t h in v i v o a n d i-n -v i t r o l e v e l s . A t t h e in v i v o l e v e l s , t h e f o u r c h a l c o n e s were o r a l l y -. .. a d m i n i s t e r e d a t h i g h t o l e r a b l e d o s e r e g i m e n w h i c h d i d n o t d i s p l a y any o b j e c t i v e s i g n s of t o x i c i t y t o mice. The compounds were a d m i n i s t e r e d t o mice w i t h S c h i s t o s o m a m a n s o n i i n f e c t i o n of 7-week-old d u r a t i o n as w e l l as t o h e a l t h y mice a t a d o s e l e v e l o f 1000 mg/kg f o r 7 c o n s e c u t - i v e d a y s . P r a z i q u a n t e l was a d m i n i s t e r e d t o mice a t a s i n g l e 7- o r a l d o s e of 685 mg/kg. The i n t e r a c t i o n o f t h e s e compounds b,. w i t h s c h i s t o s o m i a s i s mansoni was e v a l u a t e d by m o n i t o r i n g t h e i r e f f e c t on ( 1 ) worm b u r d e n ( 2 ) t o t a l l i v e r t i s s u e egg c o u n t and oogram (3) r e l a t i v e w e i g h t and s p l e e n w e i g h t ( 4 ) l i v e r e n z y m a t i c p r o f i l e of a r g i n a s e , o r n i t h i n e carbamopltransferase, a l k a l i n e phospha t a s e , c h o l i n e s t e r a s e , 5lnucleot i d a s e , a l a n i n e a m i n o t r a n s f e r a s e and a s p a r t a t e aiminotransf e r a s e . A t t h e -i n -v i t r o l e v e l , t h e f o u r c h a l c o n e s a s w e l l a s p r a z i q u a n t e l h a v e been t h e s u b j e c t o f i n v e s t i g a t i o n t o e v a l u a t e t h e i r a n t i s c h i s t o s o m a l . a c t i v i t y . The e f f e c t o f t h e s e compounds on a d e n o s i n e t r i p h o s p h a t a s e and chol i n e s t e r a s e w e r e s t u d i e d i n h omo g e n a t e s o f -S. ma n s o n i worms. I n summary, t h e r e s u l t s of t h e s t u d y h a v e r e v e a l e d t h e f o l l o w i n g : 1. A t t h e d o s e r e g i m e n s a d m i n i s t e r e d , t h e f o u r c h a l c o n e s showed e q u a l l y s l i g h t s u b j e c t i v e t o x i c i t y i n n o r m a l h e a l t h y mice as m a n i f e s t e d by e n z y m a t i c d y s f u n c t i o n of t h e l i v e r a l a n i n e a m i n o t r a n s f e r a s e and a s p a r t a t e a m i n o t r a n s f e r a s e . 2. A d m i n i s t r a t i o n o f t h e c h a l c o n e s t o mice i n f e c t e d w i t h -S . ma n s o n i d i d n o t d i s p l a y a n y s c h i s t o s o m i c i d a l a c t i v i t y where n e i t h e r r e d u c t i o n i n t h e worm b u r d e n n o r c h a n g e s i n t h e oogram w a s o b s e r v e d . F u r t h e r m o r e , no a l l e v i a t i o n of t h e i n v o l v e m e n t s due t o S. mansoni i n f e c t i o n was n o t i c e d a t e i t h e r t h e o b j e c t i v e ( o r g a n s s t a t e ) o r s u b -j e c t i v e ( l i v e r e n z y m a t i c p r o f i l e ) l e v e l s f o l l o w i n gt h e compound a d m i n i s t r a t i o n . I n s t e a d , t h e c h a l c o n e s ,i n p a r t i c u l a r chalcone-1 a n d c h a l c o n e - 2 , f u r t h e r e dt h e e n z y m a t i c d i s t u r b a n c e s c a u s e d by -S. -mans-oni i n f e c t i o nas e v i d e n c e d by t h e a d d i t i v e d i m i n u t i o n in o r n i t h i n ecarbam~yltransferase, a l a n i n e a m i n o t r a n s f e r a s e , anda s p a r t a t e a m i n o t r a n s f e r a s e i n a d d i t i o n t o a u g m e n t a t i o no f a l k a l i n e p h o s p h a t a s e a c t i v i t i e s o f bilharzial l i v e r .3 . A d m i n i s t r a t i o n o f e i t h e r chalcone-1 o r c h a l c o n e - 2 t o-S . m a n s o n i - i n f e c t e d mice c a u s e d ma r k e d e l e v a t i o n i nt h e l i v e r t i s s u e e g g c o u n t . A c c o r d i n g l y , t h e r e l a t i v e l yh i g h e r t o x i c i t y o f t h e s e two compounds c o m p a r e d w i t ht h e o t h e r two c o u l d be a t t r i b u t e d t o t h e i r p o s s i b l ee x c i t a t o r y e f f e c t on t h e e g g l a y i n g c a p a c i t y o f t h eworm t h a t c o n s e q u e n t l y l e a d s t o i n c r e a s e d number o fe g g s t r a p p e d by t h e l i v e r . T h i s s i t u a t i o n raises t h ef r e q u e n c y o f t h e d e v e l o p m e n t of i n f l a m m a t o r y granulomatousr e a c t i o n s a r o u n d d e p o s i t e d o v a h e n c e i n c r e a s e dt h e s e v e r i t y o f t h e i n v o l v e m e n t s d u e t o t h e i n f e c t i o n .4. A l t h o u g h p r a z i q u a n t e l d i d n o t c a u s e c o m p l e t e c u r e a tt h e d o s e l e v e l u s e d (685 mg/kg), i t p r o d u c e d markeda l l e v i a t i o n b o t h , o b j e c t i v e l y as mani fested by c a u s i n gt h e l i v e r a n d s p l e e n t o a t t a i n i t s n o r m a l s i z e a n ds u b j e c t i v e l y by n e a r l y n o r m a l i z a t i o n of e n z y m a t i ca c t i v i t i e s i n d i c a t i v e o f l i v e r damage.5 . -I n v-iteo s t u d i e s c o n f i r m e d t h a t n o n e o f t h e f o u r chalco n e s - h a s s c h i s t o s o m i c i d a l a c t i v i t y w h e r e t h e y d i dn o t d i s p l a y marked i n h i b i t o r y e f f e c t on e i t h e r a d e n o s i n et r i p h o s p h a t a s e o r a c e t y l c h o l i n e s t e r a s e a c t i v i t i e s o ft h e whole worm homogenate e v e n a t h i g h c o n c e n t r a t i o n . P r a z i q u a n t e l , on t h e o t h e r h a n d , showed marked i n h i b - i t o r y e f f e c t on t h e worm a d e n o s i n e t r i p h o s p h a t a s e , b u t n o t a c e t y l c h o l i n e s t e r a s e a c t i v i t y . The i n h i b i t o r y e f f e c t of p r a z i q u a n t e l w a s r e c o r d e d e v e n a t low d r u g c o n c e n t r a t i o n .
