Abstract

Casein micel.les of buffalo ’s milk were f r a c t ionated by centrifugation into large (PI ) , medium small (PIII) micelles and FIV which contained the remaining casein non-sedimented a t 10,500 G f o r >.6O min The prepared micelles were q u a l i t a t i v e l y analysed f o r i t s e l e c t r o p h o r e t i c p a t t e r n s on polyacrylamide gel e lectropharesis and f o r chromatographic behaviour on mi ce cellulose column chromatography i n the presence of t r i s - c h l o r i d e urea b u f f e r ’p8~.6 , and on sephadex .G-150 column i n the presence of t r i s - c i t r a t e urea buffer at the same pH respectively. Also, the casein micelles were q u a n t i t a t i v e l y analysed f o r its content from the major casein c o n s t i t u e n t s 8 -, K-9 B- and d s- c a s e i n by ~~~E’-cellulocsoelu mn chromatography’. K-casein was iiolated from t h e casein micelles of d i f f e r e n t s i z e s and characterized by polyacrylamide gel electrophoresis. S i a l i c acid and hexose contents were determined i n casein micellar f r a c t i o n s , and i n K-casein i s o l - ated from these micelles, whereas, the a b i l i t y of the isolated K-casein to s t a b i l i z e 0(s-casein was tested i n t h e , p resence of d i f f e r e n t concent rat ions of ~s++i n solution.
