Abstract

The crude extract of the toxin from the skin of the Red Sea fish Arothron hispidus was administered intraperitoneally in albino ratq at three different doses of 140, 160 and 165 &kg. The present study attempts to trace its effects to metabolic disorders i n brqin , liver and blood of treated albino rate. The eelected metabolites to be studied were carbohydrates, totel lipida and total free amino acids, i n addition to the activity of certain enegmes for oonvenience aa glucose -6- phosphatase, succinic dehydrogenaee snd lipase. The carbohydrates levels were greatly affected by tetrodotoxin. Generally, the glycogen content of both liver and brain was found to decrease, as the doae level of tetrodotoxin increased. While blood glucose level increaaed as the dose of the toxin increased. from the observed data, the total lipid contentsof both l i v e r and blood was found to increa~ea s the dose of tetrodotoxin increased. While the lipid content of brain was found to decrease by increasing the dose of tetrodotoxin. The levels of the t o t a l f r e e amino-acids was less affected by treatment with tetrodotoxin, since it wss clear that, the effect of this toxin on the levels of total free amino-acida of the atudied organa showed limited decreasee i n their contents. Glucose-6-phosphata~e: Generally, indicated marked increase i n the enzyme with the increase of tetrodotoxin dose. the result obtained activity in the liver, Succinic dehydrogenaae: Measurement 8 of the activity of succinic dehydrogenase showed moderate decrease i n the enzyme activityas a result of treatment with tetrodotoxin, in all the atudied organa. Measurements of the lipase a c t i v i t y i n blood and liver showed increases i n the enzyme activity as the dose of the toxin increased. Finally, it is concluded that:- - Tetrodotoxin induces its action by di~rupting the metabolic pathways of both carbohydrate and lipid metabolism via activation of lipase and retardation of Kreb’s cycle aa affected by inhi bition of succinic dehydrogeaaae activity. - The decrease recorded i n glycogen content of liver and the concomitant increase i n the blood glucose level might be attributed to the recorded increaee i n glucose -6- phosphatase activity due to tetrodotoxication. This may supply potential energy (glucose) to organa that capable of getting their needs of energy via the anaerobic oxidation and not the aerobic one, reflected i n the present work in retarded succinic dehydrogenase activity. - The limited decreaaes of the t o t a l free amino-acid in the studied organa might be attributed to the toxic effect of tetrodotoxin on the active transport mechanism of aminoacids into the cells, as affected byimpaxing energy. supply (glucose) to these organs which i a responsable for transport process.
