Abstract

This study was aimed to, early diagnosis of liver fibrosis induced by CCl4 in guinea pigs and evaluate hepatoprotective effect of pentoxifylline and fucoidan through detection of gene expression of TGFβ-1 by RT-PCR, expression of α SMA by Immunohistochemistery, alpha fetoprotein estimation, oxidative stress reaction through estimation of MDA, SOD ,GSH and CAT enzymes, biochemical parameters as well as some hematological prameters and histopathological studies by routine and especial stains. The experiment was conducted on 80 Guinea pigs of 1-2 month old. After 15 day Period of acclimatization in cages condition, Guinea pigs were divided as follow: Group I (10) take olive oil (control),Group II (10) treated with PTX and olive oil (PTX) , Group III (10) treated with fucoidan and olive oil (F),Group IV (10) treated with CCl4 (CCl4), Group V pretreated with PTX for one week before CCl4 administration then treated with CCl4 and PTX (protective PTX treated group).Group VI (10) treated with CCl4 for 8 weeks and start treatment with PTX at 4th week of CCl4 treatment (therapeutic PTX treated group).Group VII (10) pretreated with fucoidan for one week before CCl4 administration then treated with CCl4 and fucoidan (protective fucoidan treated group).Group VIII (10) treated with CCl4 for 8 weeks and start treatment with fucoidan at 4th week of CCl4 treatment (therapeutic fucoidan treated group). The CCl4 was orally administrated at dose 1 ml/kg bw (20% in olive oil) twice a week for 8 weeks in CCl4 treated groups as well as olive oil orally administrated at dose 1ml/kg bw for non CCl4 treated groups. While PTX and fucoidan were administrated orally at dose 20 mg/kg bw/day and 200 mg/kg bw/day respectively allover the experiment. Our result revealed that both of fucoidan and PTX reduced gene expression of TGFβ-1 and expression of α SMA especially protective treatment. Also protective PTX treatment corrected elevated ALT, T.Bili and GSH induced by CCl4 treatment. Also improved hypoglycemia induced by CCl4 treatment . We concluded that PTX has mild hepatoprotective effect than fucoidan especially in the protective treatment.
