Abstract In this study isolation, purification and transplantatiojn of rat pancreatic islets were carried out for the treatment of diabetic rats. The recipient rats were made diabetic by a single intravenous injection of streptozotocin. Isolation of rat islets were done by injection of collagenase type XI through the bile duct. This procedure causes the complete destruction of pancreatic exocrine tissue, as well as mechanical separation and digestion of the interstitial tissue between the islets and the exocrine tissue, with the islets being preserved selectively intact. These islets were purified by ficoll density gradient and then hand picking under stereomicroscope. |
