Abstract The main objective of the present study was: the in vitro selection of drought tolerant sunflower callus line as well as studying the characteristics of this selected line in a trial to understand the mechanism(s) of drought tolerance on the cellular level. - The first experiment was conducted to identify the optimum conditions for sunflower callus formation, and to study its regenerative capability. The best growth of sunflower callus was achieved using hypocotyl as an explant on MS medium supplemented with 1.0 mg/L NAA and 1.0 mg/L KN incubated at 28oC under 16 hrs photoperiods. - No root or shoot induction was observed on callus grown on MS medium supplemented with NAA and KN. However, substituting KN with BAP succeeded to initiate very poor microshoots but not roots. . |
