Abstract

Nitro Oxide and Antioxidants This investigation was divided into two major parts:
A) In-vivo study:
In this study, aminoguanidine (AG), nicotinamide (Nic), silymarin, vitamin E (Vit.E) and glimepiride (Glim) were administered to normal diabetic, rats and as a pretreatment before induction of diabetes in rats. Different parameters were recorded after 10 and 45 days of (STZ) injection in pretreated rats. These parameters include blood glucsoe, serum insulin, beta-cell function, insulin resistance, liver glycogen, glycated haemoglobin, serum fructosamine, serum (LDH) and (MDA) levels. Histopathological examination of pancreas, liver and kidney was performed in various groups.
B) In-vitro study: Pancreas was isolated from pretreated rats after 45 days of STZ injection and perfused with 3 mM and 16.7 mM glucose. Insulin out put was measured at different time intervals and areas under the insulin secretion-time curves were calculated.
The following results were obtained:
1- Injection of STZ in a dose of 45 mg/kg induced a significant increase in serum glucose level, glycated haemoglobin, fructosamine, LDH, MDA levels and insulin resitance. It also reduced serum insulin, beta-cell function and liver glycogen content. Histopathological examination of pancreas revealed severe islet destruction and insulitis. In-vitro study showed the disappearnace of biphasic insulin secretion pattern in response to 16.7 mM glucose in diabetic rat pancreas. STZ also induced dramatic reduction in basal and glucose stimulated insulin secretion. It reduced the area under the insulin secretion-time curves
