Abstract

N atamycin (a member of the polyene antifungal antibiotics) is effective against a broad variety of fungi, yeasts, some protozoa and some algae. It is used for the local treatment of candidiasis (oral and vaginal), trichomoniasis and fungal keratitis. Natamycin 1s available commercially as vaginal tablets (Pimafucin®) for the treatment of vaginal candidiasis. Commonly used conventional dosage forms, such as creams, foams, tablets, gels, rinses, mouthwashes, irrigations, or pessaries suffer from many disadvantages such as their relatively short residence time at the site of action. Moreover, multiple daily doses are often required, leading to systemic adverse effects and poor patient compliance. In recent years, significant interest has been shown in the development of bioadhesive dosage fonns for the delivery of drugs to the buccal and vaginal cavities for the treatment of different diseases. The present work has aimed to formulate natamycin •as buccal and vaginal bioadhesive tablets in order to prolong the residence time of the drug at the site of action and hence improve its therapeutic efficacy and enhance patient compliance due to the reduced frequency of application. So the drawbacks of the conventional dosage forms can be overcome The work in this thesis has been classified into four chapters.
