Abstract

The present study was carried out at invitro fertilization (IVF) Laboratory, International Livestock Management Training Center Sakha, Kafr El-Sheikh District, belonging to the Animal Production Research Institute, Ministry of Agriculture Egypt in cooperation with Poultry Production Department during the period from September 2005 to June 2006. .The results obtained from the analysis of the data could be summarized as follows: 1- Average absolute weight of the ovaries was heavier (0.412 vs. 0.263 g, P>0.01), ovarian weight relative to LBW (0.09 vs. 0.15 g/kg LBW, P>0.01) and ovarian length (1.313 vs. 1.554 cm, P>0.05) in does recovery embryo time after 72 than 24 hrs of mating. . 2- The differences in number of follicles/ovary did not differ significantly between does recovery embryo time after 72 or 24 hrs of mating. .3- Number of CL/ovary increased (P>0.001) in does recovery embryo time after 72 than 24 hrs of mating (12.8 vs. 16.4/doe ).. 4-There were insignificant differences in averages of absolute weight, relative weight to LBW and length of both oviduct and uterine between does recovery embryo time after 72 or 24 hrs of mating. 5- recovery rate was higher (P>0.01) for does recovery embryo time after 24 h (67.7%) than those recovery embryo time after 72 h ( 53%) of mating. 6- Survival rate of embryos cultured was more on the 2nd and 3rd day for embryos recovered after 24 (95.9 and 83.8%) than 72 hrs (90.4 and 66.3%) of mating, respectively. 7- Survival rate was nearly similar on the 2nd day of culture TCM-199 and HAM F-10 (92.6 vs. 93.4%, respectively), while survival rate of embryos was slightly lower in TCM-199 than in Ham medium (74.1 vs.75.0%). 8- Embryos recovered after 24 hrs of mating and in vitro cultured in Ham showed the highest survival rate on the 2nd and 3rd day of culture (97.1 and 85.7%) versus the lowest values for those recovered after 72 hrs of mating and cultured in Ham medium (90.2 and 65.8%), respectively. 9- Thickness of mucin coat increased (P<0.05) and zona pellucida decreased (P<0.05) with vitrification as compared to slow freezing method. However, diameter of interzonal and embryo with its coverings were not affected significantly by preservation method. 10- Thickness of mucin coat and diameter of embryo with its coverings increased (P<0.05) with TCM than Ham, however, thickness of zona pellucida and diameter of interzonal were not affected significantly by type of culture media. 11-Thickness of mucin coat and zona pellucida as well as diameter of embryos with its coverings were higher (P<0.05) with vitrification than slow freezing and with Ham than TCM. However, diameter of interzonal was not affected significantly by preservation method or type of medium. 12- Thickness of mucin coat as well as total diameter of interzonal and embryo with its coverings increased (P<0.05), while thickness of zona pellucida decreased (P<0.05) by advancing culture day after thawing for three days. 13- 13- percentage of live born insignificantly increased for vitrified than slow frozen embryo 50.0%) versos 46.6% 1- 2 14- rabbit does should be superovulated by PMSG and HCG and recovery embryo time after 24 h of mating when embryos at early stage are needed for banking females of high genetic merit, and recovery embryo time after 72 h of mating if embryos at morula stage were needed for embryo transfer. 2- 3- 15-Cryopreservation of rabbit embryos in morulae stage by vitrification was better than slow freezing for embryo transfer and high live born. 4-
