Abstract

This study was conducted on potato crop cv. Spunta in the tissue culture laboratory of the Horticulture Department, Faculty of Agriculture, Mansoura University during the period from 2002 to 2005, aiming to investigate the effect of cytokinins, auxins, sucrose and salinity stress on micropropagation and microtuberization of Solanum tuberosum L. The experiment included 4 parts as following, effect of BAP, KIN, NAA and their interaction on shoot proliferation of potato by using 5 levels of BAP or KIN (0.0, 0.1, 0.2, 0.5, 1.0 mg/L) with (0.0, 0.1 mg/L) NAA, effect of different concentrations of sucrose with 4 levels (30, 60, 80, 100 gm/), BAP with 7 levels (0.0, 0.1, 0.2, 0.3, 0.4, 0.5, 0.6 mg/L), 2,4­D, IBA and NAA with 4 levels (0.0, 0.5, 1.0, 1.5 mg/L) on microtuberization of potato, effect of different concentrations of NaCl with 8 levels (0.0, 0.5, 1.0, 2.0, 4.0, 6.0, 8.0, 10.0 gm/L) on shoot proliferation, effect of different concentrations of NaCl with 7 levels (0.0, 0.5, 1.0, 2.0, 4.0, 6.0, 8.0 gm/L) on microtuberization of potato. The experimental design was complete randomized design. In all experiments each treatment was represented by 9 jars of three replicates (three for each), and each jar included 3 explants. In conclusion, it could be concluded that using MS supplemented with 0.2 mg/L BAP + 0.1 mg/L NAA resulted the best micropropagation method, using 2.0 mg/L BAP + 80 gm/L sucrose under dark conditions resulted the best microtuberization, using 2,4­D at 1.0 mg/L, IBA 1.5 mg/L and NAA at 1.5 mg/L were the best for microtuberization, under vitro conditions potato overcame the harmful effect of salinity stress (up to 4000 ppm) and proliferated high quality and quantity shoots and microtubers.
