Abstract

Cancer breast is the most common malignancy among females worldwide . The first step for development of the tumor may be a genetic defect and further genetic mutations accumulate in the cells to increase the malignant potential. Often these mutations occur in tumor suppressor genes such as p53. So, the objective of this study is to investigate the expression of P53 in breast cancer patients and its relation with some prognostic values. Our study was conducted on 33 female patients with primary breast carcinoma . They were compared to 10 patients with breast fibroadenoma as a control group for the evaluation of P53 in benign breast conditions. Fresh tumor samples were obtained from the patients and the controls at the time of the diagnosis to be examined by (FCM) for the expression of P53. Statistical analysis of our results showed the following: There is statistical significant increase in P53 level in the patient group compared to the control group. There is statistical significant increase in P53 level in the ductal carcinoma compared to the lobular carcinoma. There is statistical significant difference in P53 level among the three stages of the tumor. There is statistical significant difference in P53 level among all tumor size groups. There is statistical significant increase in P53 level in cases with positive draining lymph node metastasis compared to cases with negative draining lymph node metastasis. There is negative statistical significant correlation between the level of P53 and the age of the patients. CONCLUSION: from these results, we conclude that detection of P53 by (FCM) is rapid, objective and quantitative method . Over expression of P53 protein detected by flowcytometry is associated with more advanced and/or more aggressive tumors, increasing in frequency with disease progression and poor patient prognosis. So, it may be used as an independent prognostic factor for evaluation of patients with cancer breast. RECOMMENDATIONS: DNA sequencing of mutant P53 in patients with P53 over expression is undergoing to elucidate the type of mutation present in our region.
