Abstract

Administraiton of streptozotocin to neonatal rat resulted in significant increase in both plasma glucose and plasma insulin 6 weeks after infection. Also significant increase in plasma MDA, TNF and lipogram. Streptozotocin produces significant reduction in body weight and non significnat changes in liver function tests. Administration of subcutaneous NPH insulin produces non significant change in body weight, plasma glucose, MDA, TNF , lipogram and liver function tests while it produces significant increase in plasma insulin level. Administration of troglitazone alone produces significant body weight reduction together with significant reduction in plasma level of glucose, insulin, MDA, TNF and lipogram. However, it produce significant increase in plasma AST, non significant increase in plasma ALT. Coadiministration of troglitazone and insulin produced significant reduction in plasma level of glucose, insulin, lipogram, MDA and TNF but it produces significant increase in plasma level of ALT, nonsignificant increase in plasma ALT level. Coadministration of troglitazone and silymarin produced significant decrease in plasma AST level.In the light of the present study it could be concluded that troglitazone has an insulin sensitizing effect either alone or combined with insulin and it has valuable role in treatment of type 2 diabetes but with its possible hazardous effect of hepatotoxicity and increase serum aminotransferase level, the use of hepatoprotective drug as silymarin was beneficial but their therapeutic use in combination with thiazolidinedione drug group need further human studies.
