Abstract

Introduction: Ovarian cancer remains the most common cause of mortality from gynecologic malignancies (1) and fourth common cause of death among all malignancies in women. The prognosis of patients with ovarian cancer is related to several factors: age of the patient, stage of the disease, histological grade of the tumor, size of the residual tumor, presence or absence of P53 gene mutation and second-look status. Aim of the work: To evaluate the effect of P53 gene mutation on the biologic features of ovarian cancer Study Design: The study included 40 cases of ovarian cancer and 10 control cases with normal ovaries. P53 study was done by IHC and PCR - SSCP techniques, and the cases were followed up by ultrasound, CT scan and CA125 level . Results: P53 overexpression was positive in 27 cases (67.5%). P53 gene mutation was detected by PCR-SSCP technique in 23 cases (57.5%). The general concordance between the two techniques in detection of p53 alteration was 85%. All control cases were negative for p53 alteration. P53 gene mutation was found to be associated with: advanced stage of the disease, poor grade of differentiation , less feasibility of cytoreductive surgery , more recurrence rate , a shorter disease-free interval and a shorter mean survival time Conclusion: It can be concluded from this study that p53 gene alteration adversely affects the biological features of ovarian cancer.
