Abstract

The aim of the work was (Studies on Thermostable Microbes­producing for Lipase). The present studies were devoted to study the following objectives: Comparison of the potentialities of thermostable bacteria and fungi producing of lipase, Optimization the production of lipases of the most active thermophilic bacteria and fungi, Immobilization of crude lipase using sodium alginate and studying the properties of the immobilized lipase in comparison with free enzyme and Studying the hydrolysis of oils and fats by the lipases. The results obtained can be summarized in the followings: Section I: Comparison of the potentialities of thermostable bacteria and fungi producing of lipase: The lipase production by seven bacterial strains and six fungal strains was evaluated. Bacillus stearothermophilus and Bacillus licheniformis were the most active among the lipolytic bacterial strains. Aspergillus fumigatus and Aspergillus nidulans were the most active lipase producers among test fungi. Section II: Factors affecting lipase production: The factors affecting lipase production were studies including: time course, initial pH, temperature, carbon source, carbon concentration, nitrogen source, lipids source, oil concentration, fatty acids and shaking. Section III: Characteristics of immobilized lipase: The lipases produced by Bacillus licheniformis and Aspergillus nidulans were isolated from the culture supernatants by cold acetone then immobilized in alginate beads.
