Abstract Cisplatin (CP) is a widely used chemotherapeutic agent used to treat solid tumors such as head, neck,ovarian, and testicular cancers. Although having a good therapeutic potential, its uses are limited due to the major and severe side effects such as nephrotoxicity,Bone marrow suppression, gastrointestinal toxicity, ototoxicity, and neuropathy.Nephrotoxicity is the main dose-limiting adverse effect in 25%–35% of patients treated with even a single dose of CP.Amifostine is the only FDA-approved drug to prevent CP-induced nephrotoxicity. However, the cytoprotective effect of Amifostine is not consistently demonstrated, with multiple reports showing evidence of toxicity despite administration at optimal doses. No treatment strategies are currently available for the prevention of CP-induced nephrotoxicity;only hydration therapy with the administration of diuretics is used as a preventive measure. Thus,there is an urgent need for the development of effective and safe alternative therapies that can be used to prevent CP-induced nephrotoxicity. |
