Abstract

Hematopoietic stem cell transplantation (HSCT) is the only curative treatment of different acquired and inborn hematological diseases in children. The main goal of post-transplantation chimerism monitoring in HSCT is to predict negative events, in order to intervene with appropriate therapy and improve the probability of long-term disease free survival. We conducted our study on 9 recipients (six thalassemia and three aplastic anemia patients) and their donors. We used multiplex PCR based short tandem repeats (STR) analysis for quantifying the relative amounts of donor and recipient cells present in the peripheral blood sample to determine if engraftment has taken place or not and the state of chimerism. Seven (77.8%) of our patients showed complete chimerism (CC), one (11.1%) showed mixed chimerism (MC) and one (11.1%) showed autologous recovery (AR). Six patients who showed CC are disease-free survivors. One patient with CC had graft failure. One patient who showed MC died. The patient who showed AR had graft failure. Moreover, we compared our results with the other methods used for detection of chimerism in these patients
