Abstract

Title: Long non-coding RNA RMRP as diagnostic marker in Egyptian patients with vitiligo.
Vitiligo is an acquired pigmentary skin disorder of unknown origin characterized by the absence of pigmentary cells from the epidermis that results in white macules and patches on the body.
Long non coding RNAs (lncRNAs) are defined as ncRNAs that longer the intergenic stretches or overlapping antisense transcripts of protein coding than 200 nucleotides without the capacity of coding proteins, located within genes. There is growing evidence that lncRNAs participate in various biological events, and many studies have suggested that dysregulation of lncRNAs are involved in the pathogenesis of autoimmune and inflammatory diseases.
Our study included 20 patients with vitiligo and 20 healthy controls. .
Blood samples were taken from patients and examined for LncRNA RMRP by quantitative real time –PCR and 3 mm Tissue biopsies were taken and examined for LncRNA RMRP by quantitative real time -PCR.
Our results showed that serum and tissue RMRP is significantly could be diagnostic marker for vitiligo (p value<0.001, AUC=1) at cut off value 1.11, 1.6 sensitivity 99%, specificity 99% and accuracy 99%.
We suggest that up regulation of lnc RNA RMRP may have a role in vitiligo pathogenesis through blockage of cell cycle progression and induction of melanocyte apoptosis and increased levels of Th17 and high IL17.
