Abstract Aptamers are believed to be advantageous chemical analogs of antibodies that can be perfectly used as detecting agents for a certain pathogen due to their superior stability, sensitivity and specificity. Herein, specific aptamers were generated and selected through an improved cell-SELEX methodology that combined conventional and toggle cell-SELEX. This methodology was designed to avoid the common pitfalls of current methods using different strategies. The enhanced method was applied to select candidate aptamers for the three classic Brucella species circulating in Egypt as a proof of concept. Brucellae were perfect nominees for their global zoonotic importance and the urge for a novel direct diagnostic tool. The selected aptamer, BR8-15, revealed high specificity for the three Brucella species with high binding affinity in the nanomolar range and 101 CFU/mL detection limit. |
