Abstract

This study aims to investigate the effect of Passion fruit (Passiflora edulis) for the oxidative stress induced by potassium bromate in male albino rats.
Thirty six white male albino rats, Sprague Dawely strain, 2-week age, weighting (120-160 g) will be used.
The rats will be acclimatized for a week under laboratory conditions before commencement of the experiments.
KBrO3 will be used as single intraperitoneal injection (125 mg/kg B.Wt).
This group was further divided into the following subgroups:
• group (1): (Control ” – ”) rats (n=6) will be fed on basal diet only.
• group (2): (Control ” + ”) rats (n=6) will be treated with KBrO3 solution then will be fed on basal diet only.
• group (3): Rats (n=6) will be treated with KBrO3 then will be fed on basal diet containing 2.5% of Passion fruit pulp powder.
• group (4): Rats (n=6) will be treated with KBrO3 solution then will be fed basal diet containing 5% of Passion fruit pulp powder.
• group (5): Rats (n=6) will be treated with KBrO3 solution then will be fed on basal diet containing 2.5% of Passion fruit peels powder.
• group (6): Rats (n=6) will be treated with KBrO3 solution then will be fed on basal diet containing 5% of Passion fruit peels powder.
At the end of experiment (4weeks), the blood samples were collected after 12 hours fasting and serum was separated for determination of:
Lipid profile , total cholesterol (TC),triglycerides (TG),high density lipoprotein cholesterol (HDLc), low density lipoprotein cholesterol (LDLc), very low density lipoprotein cholesterol (VLDLc) , Urea , creatinine, Uric Acid, Glotamic Oxaloacetic transaminase (GOT) , Glotamic pyruvic transaminase (GPT) , Alkaline phosphatase (ALP), antioxidant enzymes (CAT,GOX and SOD), total protein, albumin, globulin and serum glucose .
At the same time ,the organs : kidney and liver were removed ,washed in solution ,dried by filter paper , weighted .Liver and kidney stored frozen in formalin solution 10% for histopathological examinations.
Statistical analysis
The data were statistically analyzed using a computerized costate program by one way ANOVA. The results are presented as mean ± SD. Differences between treatments at (p≤0.05) were considered significant.
The results can revealed as following:
1. There were significant decreases in BWG, FI and FER of rats injected with KBrO3 of control (+) group, while rats injected by KBrO3 then fed on all tested plants had significant increases in BWG, FI and FER when compared to control (+).
2. Rats fed on groups 4 (Passion fruit pulp 5%) showed the best results as for FI for rats when compared to control (+).
3. Rats fed on group 4 (Passion fruit pulp 5%) recorded the best treatment of BWG and best treatment of FER when compared to control (+).
4. There were significant decreases in relative liver, spleen, lungs, heart and kidneys weights of rats injected with KBrO3 as compared to control (-) group.
5. Rats fed on group 4 (Passion fruit pulp 5%) showed the highest mean value of liver, kidney, heart, lungs and spleen when compared to control (+).
6. Rats treated with KBrO3 and fed on group 4 (Passion fruit pulp 5%) showing the best treatment of serum glucose as compared with control (+) group.
7. Rats treated with KBrO3 significantly increases in the serum total cholesterol (TC) and triglycerides (TG), which were decreased when rats treated with KBrO3 and fed on diet containing the Passion fruit. Best group of serum TC and TG was recorded for group 4 (Passion fruit pulp 5%) when compared to control (+).
8. The best serum of HDL was found in rats was found in group 4 (Passion fruit pulp 5%) when compared to control (+).
9. LDL & VLDL level increased markedly in the serum of rats treated with KBrO3. Feeding on Passion fruit pulp and peels decreased the levels. The best group of LDL and VLDL was recorded for group 4 (Passion fruit pulp 5%) when compared to control (+).
10. Rats treated with KBrO3 was damaging to the liver function as indicated by the rise of AST, ALT & ALP. Nevertheless feeding on Passion fruit pulp and peels corrected the change of AST, ALT and ALP. The best serum AST, ALT and ALP was recorded for group 4 (Passion fruit pulp 5%) when compared to control (+).
11. Rats treated with KBrO3 affected the kidneys function giving rise to serum urea, creatinine and uric acid while feeding on Passion fruit pulp and peels reversed this change.
12. The best serum of urea, creatinine and uric acid was noticed for group 4 (Passion fruit pulp 5%) when compared to control (+).
13. Rats treated with KBrO3 decreased in antioxidant enzymes (CAT, GPX and SOD) while feeding on Passion fruit pulp and peels increased in antioxidant enzymes.
14. The best serum of CAT, GPX and SOD was noticed for group 4 (Passion fruit pulp 5%) when compared to control (+).
15. The best serum of total protein and albumin was noticed for group 4 (Passion fruit pulp 5%) when compared to control (+).
16. The best serum of globulin was noticed for group 4 and 5 (Passion fruit pulp 5%& Passion fruit peels 2.5%) when compared to control (+).
