Abstract Psoriasis is a common chronic autoimmune and inflammatory skin disease in which both genetic and environmental factors have a critical role. The most characteristic lesion is red, scaly sharply demarcated, indurated plaques over extensor surfaces and scalp. It affects 2.7% of the world’s population. At the onset of psoriasis, special DCs in the epidermis and dermis are activated among other effects, these cells produce the messenger substances e.g TNF-α which in turn promote the development of T helper (Th)1, and Th17 cells. These T cells secrete mediators that contribute to the vascular and epidermal changes of psoriasis. The hCYP1A1 is an enzyme of biomedical and toxicological interest, which is expressed in extrahepatic tissues, catalyzes the biotransformation of polycyclic aryl hydrocarbons (PAHs), aromatic amines, and polychlorinated biphenyls into polar compounds, which can be conjugated to soluble compounds suitable for excretion by urine or bile. Nevertheless, under specific circumstances, this enzyme catalyzes the bioactivation of compounds capable of reacting with macromolecules, such as DNA, leading to the start of mutagenic process. Every day, we are exposed to compounds that are substrates of CYP1A1, through environmental pollution, food, and, particularly, cigarette smoke. The importance of this protein in chemical carcinogenesis induced by PAHs has been demonstrated in CYP1 knockout mice, in which the lack of this protein shows less formation of adducts PAH-DNA. The aim of this study was to evaluate therole of cyp4501a1 gene polymorphism in the pathogensis of psoriasis and their relation to the disease severity. This case-control study included 45 unrelated patients having psoriasis, in addition to 45 age and sex apparently healthy volunteers as acontrol group. Cases of psoriasis were selected from Dermatology Outpatient Clinic, Menoufia University Hospital during the period from to while the control subjects were selected from those attending Plastic Surgery Department.
