Abstract

The probiotic bacteria, prebiotics and their combinations established positive impact on the intestinal microflora. So, the aim of this study was screening for biological activity of probiotics bacteria (the anticancer, antiviral, antimicrobial and lipidemic profile) under the effect of probiotic bacteria in sole form and its combination with prebiotics, as synbiotics and investigated the promotion and benefit of this combination. Anticancer activity tests were performed against three types of cancer cell lines Hepg2 (Liver cancer), A549 (Lung cancer), and Caco2 (Colon cancer). Lactobacillus acidophilus was used as probiotic in supernatant and cell sonicate state, while grape seeds and licorice root extracts were used as prebiotic. Data showed that anticancer was cell type and formulated Pre/Probiotics formula dependent. Combined form of probiotics and prebiotics showed a much-reduced IC50 and cell cycle accompanied with elevated early and late apoptosis compared with control, also caspase3 gene expression was significantly elevated. Antiviral activity tested against HSV1 and VSV. Assessment of the antiviral activity against HSV1 using L. acidophilus cell sonicate. L. acidophilus supernatant, L. acidophilus mix, anise, L. plantarum cell sonicate, L. plantarum supernatant and L. plantarum mix showed higher antiviral potential. While L. acidophilus mix, L. rhamnosus supernatant, anise, L. plantarum cells, L. plantarum supernatant and L. plantarum mix showed a promising antiviral potential against VSV. The Hyperlipidemia management recorded data showed that probiotics compound administration significantly decreased the total serum cholesterol and triglycerides, in addition significant increase of serum antioxidant potential. The antimicrobial activity against Staphylococcus aureus, and Pseudomonas aeruginosa pathogen strains was evaluated using both in vitro and in vivo methods. In vitro studies suggested that tested treatments have an inhibitory effect against both pathogens.The two pathogens were sensitive toLawsonia inermis (Henna) In vivo study. The administration of the LAB to mice repressed the expressions of TNF-alpha, as well as IL-6. Meanwhile WBC decreased to a remarkable value almost near control in treated groups.
