Abstract

Thioacetamide (TAA) has been used extensively in
the development of animal models of acute liver injury.
Tinospora cordifolia belongs to the family
Menispermaceae. The present study provides the evidence
indicating the protective effect of the extract prepared from
the dried leaves of Tinospora cordifolia on isolated rat
hepatocytes and an animal model of hepatotoxicity.
In the present study rat hepatocyte were isolated and
treated with different doses of Tinospora cordifolia leaves
extract. The multiple concentrations of ethanolic extract of
tested plant was used and The maximum dose has No
Observed Adverse Effect Level (NOAEL) equals (15.6
μg/g cell) were calculated from dose-response curve. The
extract exhibited no significant cytotoxic activity against
the isolated rat hepatocytes achieving a weak inhibitory
activity under these experimental conditions with IC50 = >
500 μg/ml. Quantification of oxidative stress parameters in
rat hepatocytes treated with Tinospora extract at dose (15.6
μg/g cell) . Untreated cells were used to quantify basal
oxidation. The concentration of MDA was significantly
lower (P<0.05) in TC-extract treated hepatocyte, as
compared to control untreated.
