Lornoxicam is an extremely potent non-steroidal anti-inflammatory drug belongs to the oxicams; it shows analgesic effect and widely used in management of postoperative pain associated with different surgeries. However, lornoxicam shows a distinct pH-dependent solubility characterized by poor solubility in low pH conditions present in the stomach which consequently leads to delay in its analgesic effect and increase its gastric side effect. Additionally, its usefulness is limited by its short duration of action due to its short half-life and rapid elimination. Hence it requires repeated daily administration to achieve long lasting and constant pain relief. Various techniques are used to prepare tablets; among them blister-molding method can be used (with utilizing different excipients; wetting agent, solubilizers, basic pH-modifiers and release-modification polymers) to prepare lornoxicam tablets with improved solubility and immediate/or controlled drug profile to enhance pharmacological effects and reduce potential gastric side effects. In addition to blister-molding method, hot-fusion followed by compression using Compritol® 888 ATO can successfully be used to prepare lornoxicam controlled-release tablets unlike direct compression and wet granulation methods.
