Abstract The efficacy of different vaccines, for Salmonella typhi, including Vi capsular polysaccharide (Vi - CPS), outer membrane protein (OMP) fraction, killed whole cell and the traditionally heat killed vaccine were compared. The level of antibodies after immunization of mice was estimated using the indirect whole cell ELISA technique; the whole cells as ’’coating antigen’’ were used and the sensitivity of the assay was enhanced significantly by in - situ heat treatment of the coating antigen rather than pre - heated coating antigen. Immunizing doses of Vi capsular polysaccharide, 2 and 4 æg given subcutaneously (S.C) and 4 æg given intraperitoneal (I.P), showed significant immune response. However, the response to these doses was almost similar without significant difference. Therefore, a subcutaneous dose of 2 æg is recommended. In general, the highest response was obtained using the OMP but its protection was reported to be reduced post infection with Vi - expressing Salmonella |
