Abstract The target of this study was to detect the genetic polymorphisms of some functional genes related to meat quality (DGAT1, FABP4, TG, and TCAP) in buffalo reared in Egypt. Allele identification was done by characterizing the PCR products using the PCR-RFLPs and gene amplification was digested using specific restriction endonuclease enzymes. The single nucleotide polymorphisms (SNPs) were identified by sequencing each PCR product after the purification process. Protein translation indicated the synonymous and non-synonymous SNPs and the 3D tertiary structure of selected genes peptides, prediction, and the effects of amino acids substitution on the protein structure were analyzed. The results showed different SNPs in Egyptian buffalo tested genes: one nsSNP was detected in DGAT1 and caused amino acid substitution in DGAT1 polypeptide that was predicted to be neutral and located in the coiled region of the protein. Also, two nsSNPs were discovered in FABP4 and produced two amino acid substitutions in the coiled region of the polypeptide. In TG gene prompter three SNPs were recognized. The TCAP gene showed four nsSNPs that caused four substitutions located in the α-helix region. Protein prediction analysis of amino acid substitutions showed that only the two amino acids substitution in FABP4 were conserved, while, the amino acid substitutions in DAGT1 and TCAP were non-conserved. Besides, all the substitutions have low sensitivity to mutation. |
