Abstract

Rabies vaccine is the only available treatment to prevent the development of fatal
rabies disease. A vaccine that could induce earlier neutralizing antibodies
associated with an adequate cellular immune response is required. Imject alum
and outer membrane proteins of Salmonella enterica serovar typhimurium (OMP)
were tested as adjuvants to an inactivated locally prepared Vero cells rabies
vaccine (FRV/K2). OMP combined rabies vaccine (FRV/K2 +OMP) induced
enhanced, non-delayed neutralizing antibodies relative to free FRV/K2 vaccine
and alum adsorbed vaccine (FRV/K2+alum). FRV/K2 +OMP induced
significantly higher level of IFN-ɣ that indicates activation of innate immune
cells as macrophages and natural killer cells. The percentage of CD8+ T cells
were insignificantly different post-vaccination with reference vaccine (RV), free
FRV/K2, and FRV/K2+OMP while it was significantly higher than
FRV/K2+alum group. On the contrary, the percentage of CD4+ T cells was
significantly elevated post-vaccination with free FRV/K2, and FRV/K2+alum.
Biochemical analysis and histopathological examination revealed that OMP
could be used safely as an adjuvant for the development of more effective rabies
vaccines.
