Abstract

Phytochemical investigation of Rubia tinctorum extract has led to the isolation of eight known compounds. The isolated compounds were elucidated as Scopoletin,Rubiadin,Chrysophanic acid,Alizarin,Purpurin, Nor-damnacanthal,Emodin andRutin.
Moreover,chemical profiling of R. tinctorum roots extract was performed. Methanol extract was subjected to LC-HRMS analysis to determine its chemical constituents .The obtained results indicated the prescence of 17 flavonoids and 28 anthraquinones.In addition, the semisynthetic derivatives of alizarin were prepared .The cytotoxicity of prepared compounds as well as alizarin, purpurin, methanol, chloroform, hexane and ethyl acetate extracts of R.tinctorum was evaluated against panel of cancer cell lines.The obtained results revaled that chloroform fraction of R.tinctorumwas the most selective cytotoxicity against the MCF-7, MDA-MB-231 and HepG2 cells (IC50 = 3.987, 37.9 and 22.26 µg/mL) respectively. Furthermore, the antimicrobial activity of alizarin, purpurin, methanol and chloroform extract of R.tinctorumwas determined against Gram- positive and Gram- negative bacteria. Our results indicated that they had antibacterial activity especially against Klebsiellapneumonia followed by Staphhylococcus aureus. Additionally, the invivo antidiabetic & antihyperlipidemic effect of R.tinctorum revealed that liver function enhancement, significant decreasing of body weight, improving lipid profile, normalizing hyperglycaemia, insulin resistance and hyperinsulinemia. Finally, R.tinctorum extract was evaluated for molecular docking studies that were performed to support the biological activities.
