Abstract

Cissus rotundifolia (Vitaceae) is an edible plant used traditionally in Yemen for the treatment of gastrointestinal troubles, in loss of appetite, skin diseases, burns and fever and used as antidiabetic and antimalarial. In the present work, the antidiabetic activity of the ethanolic extract prepared from the dried leaves and stems of C. rotundifolia, and its fractions; methylene chloride and n-butanol was evaluated in vitro by measuring Ü-glucosidase inhibitory activity and in vivo using STZ-induced diabetic rats. In vitro antioxidant activity was evaluated by using DPPH assay. All the tested extracts showed significant Ü-glucosidase inhibitory activity with methylene chloride fraction showed the highest activity with IC50 1.23±0.022 mg/ml compared to acarbose with IC50 0.789±0.009 mg/ml. In vivo study showed that all the tested extracts exhibited significant antidiabetic activity; with n-butanol fraction (300 mg/kg b.wt) showed the most potent activity as level of blood glucose decreased by 61.42 % compared to gliclazide (63.38 %) and insulin level was increased by 3.71 folds as compared to diabetic control group.
