Abstract

This study was preformed with the aim of investigation the expression of Midkine protein and gene in OSCC, by using immunohistochemical technicque and real time PCR. And to correlate this expression with different grades in an attempt to evaluate its role in tumor prognosis. Thirty-six specimens of paraffin blocks were investigated in this study, sixteen cases are well-differentiated OSCC, fourteen cases were poorly differentiated OSCC, and six are normal oral mucosa. from each block, one section were cut and stained with H&E, diagnosed and classified according to WHO criteria (Pindborg et al, 1997)[71]. Other sections were cut (5 microns thick) and were placed on positively charged slides (optiplus) for immunostaining procedure. In addition 25 microns from each case were placed in plastic epindorph for evaluation of midkine mRNA by using RT-PCR. For immunohistochemical examination, the primary antibody used was rabbit polyclonal antihuman Midkine antibody (US Biological, Cat. No. 211951, USA). The sections were analyzed by image analyzer computer system using the software Leica Q500 to measure the area percentage % and the immunostaining intensity (optical density) of MK in the positive cells. A total score of midkine expression for each case was obtained, student test was used to compare the mean value of total score of Midkine expression among two groups
